Dataset: "UT-GPCR005 Fluorescence anisotropy time-series of competition binding between unlabeled ligands of M1 muscarinic acetylcholine receptor and TAMRA labeled UR-MK342" Purpose: This serves as supplementary dataset to "Laasfeld, T., Bláhovcová, H., Hollo, K., Veiksina, S., Rinken, A., & Allikalt, A. Abstract: Fluorescence anisotropy time-series of competition ligand binding between unlabeled competitors of M1 muscarinic acetylcholine receptor and TAMRA labeled UR-MK342. The dataset contains kinetic traces of UR-MK259, UR-SK-75, McN-A-343, UR-SK-59, Scopolamine, Telenzepine, 4-DAMP, Carbachol, Pilocarpine, Clozapine, Pirenzepine, Atropine, Acetylcholine, Trospium, NMS, Arecoline, W84, Xanomeline and LY2119620 in competition with TAMRA labeled muscarinic acetylcholine recepotor fluorescent ligand UR-MK342. The dataset also contains non-linear regression fits of these time-series. Details of experimental setup are given in the related publication. Instructions: To read the dataset, unzip either the zip package containing the GraphPad Prism file (if you have Prism available) and/or CSV files. In case of CSV files, each ligand´s representative trace is in a separate file, all traces are in a single GraphPad Prism file. Note that in some cases the "total binding" of an experiment may contain many more replicates compared to sample due to assay format differences (multiple ligands measured in the same experiment, but 0 concetration points were done for each ligand). The first column of csv files is time since measurement start (Note: it does not always exactly match the time of mixing, usually a 30-60 s delay is expected) Also note that for each ligand the non-displacable fraction may not match the value in the paper as the variability in some cases was large and in some cases none of the experiments resulted in a value very close to the multi-experiment average. The data is organized so that column header and file name says which ligand it was (concentration information, if there is in nM or log(M) units). First columns are high concentration ligand measurements while last ones are total binding control for that experiment.
